Abstract

A novel and rapid HPLC method was developed for the determination of acrylamide (ACM), 5-Hydroxylmethylfurfural (5-HMF), patulin (PAT), and ochratoxin A (OTA) simultaneously. These four chemicals are toxic to human body which could also bring physiological effect to consumers. ACM and 5-HMF are suspected carcinogenic compounds that formed via Maillard reaction while PAT and OTA are both mycotoxin compound produced by several species of fungi. Determination of these four contaminants in coffee, tea and soy sauce which commonly consumed in Malaysia were conducted. The analytes were extracted only by using the mixture of organic solvent which was the mixture of methanol and ultrapure water with 0.1% acetic acid. After vortexed and centrifuged the samples, the four chemicals were determined by reversed-phase HPLC coupled with multiwavelength UV-Vis Diode Array Detector (DAD). The wavelength set for this method was 254nm. The method was validated for various food matrices and showed good linearity, precision and trueness. The proposed method was applied to the total of 21 samples (6 coffee samples, 9 tea samples, and 6 soy sauce samples). LC-MS/MS had been used for peak confirmation and the results were good by showing all chemicals that were detected in HPLC. However, it was found out that this HPLC method was unable to quantify the OTA as increasing concentration of the OTA cannot be well detected with the slope value of 6E-06. This HPLC method can quantify three chemicals which are ACM, 5-HMF, PAT and qualify OTA. OTA was successfully quantified using LC-MS/MS method.
