Abstract

Xanthine oxidase is an enzyme that oxidizes the xanthine and hypoxanthine to uric acid which cause hyperuricemia and gout. The example of urate-lowering agents include, which block the synthesis of uric acid in the terminal step of uric acid biosynthesis, thereby lowering the plasma uric acid concentration. However, the use of allopurinol and febuxostat can lead to some unwanted side effects such as hepatitis, nephropathy, allergic reactions and low blood count. Therefore, this experiment was carried out to determine the xanthine oxidase (XO) inhibitors from Chrysanthemum morifolium Ramat. extract by using HPLC qualitatively and quantitatively. The results showed that the ethyl acetate (EA) fraction has the highest XO inhibition of 46.43 ± 17.08%, follow by petroleum ether fraction (21.57 ± 5.38%), residual fraction (8.13 ± 1.26%) and crude extract (3.05 ± 6.18%). Therefore, the EA fraction was subjected to fractionation by gravity column chromatography. The sub-fractions, F3, F4 and F6 that showed high inhibition rate of 60-71%. After that, S3, S4 and F6were used to identify the xanthine oxidase inhibitor by using Ultra High Performance Liquid Chromatography (UHPLC) to determine the presence of quercetin, catechin and luteolin. In the presence study, quercetin was identified in F3 by using HPLC.
