Abstract

The aim of this project was to synthesize and characterize a Schiff base derivative, the thiosemicarbazone derived from thiosemicarbazide and chalcone. Antioxidant activities of the synthesized thiosemicarbazone was studied using the DPPH free radical scavenging method. In this study, the thiosemicarbazone, (Z)-2-((E)-3-(4-chlorophenyl)-1-phenylallyidene-Nmethylhydrazine- 1-carbothioamide (MT4Cl) was synthesized from 4-methyl-3- thiosemicarbazide (4M3T) and 4-chlorochalcone (4ClC) via conventional condensation reaction. The compound was characterized by Differential Scanning Calorimetry (DSC), Fourier - Transformed Infrared (FTIR) spectroscopy, Direct Injection Mass Spectrometry (DIMS), Proton (1H) and Carbon-13 (13C) Nuclear Magnetic Resonance (NMR) spectroscopy and Single Crystal X-ray Diffraction (SXRD). The structure of the compound was determined to be identical to the proposed structure. Also, antioxidant study was done via the 1, 1-diphenyl 2-picrylhydrazyl (DPPH) scavenging assay method. This method was used to evaluate the by half maximal inhibitory concentration value (IC50). Lower IC50 value corresponds to higher antioxidant activity as lower concentration value needed to inhibit the oxidation process by half. The result showed that the compound, MT4Cl did possess antioxidant activity, having IC50 of 127.70 ppm.
