Abstract

Soil bacteria are essential for most of the field of study as they have the potential biochemical properties that play a critical role in ecological and biodegradable function processes. Over the past decades, the overfill of waste in our landfill has been a serious problem facing by our society in current situations that can bring a negative effect towards the mother earth. Therefore, the main focus of this project is to isolate and identify the potential degrading bacteria from soil samples which can be effectively used for degrading kitchen waste. By using three different culture mediums, MacConkey agar, Starch agar and Skim Milk agar, that aim for different potential degrading bacteria, three main bacteria genera were obtained from the soil sample. The characteristics of bacteria were determined by conducting gram staining and biochemical test such as catalase, oxidase, coagulase, methyl-red (MR) and voges-proskauer (VP) test. By performing gram staining, gram-negative bacteria were observed from both MacConkey agar and Skim Milk agar whereas gram-positive bacteria were observed from Starch agar. Regarding biochemical characteristics, the reaction of bacteria towards the tests performed indicated that the isolates belonged to three genera, which are, Escherichia, Pseudomonas and Bacillus. In a nutshell, this study contributes with microbial isolates from soil environments with potential degrading ability in improving the biodegradable process of kitchen waste under controlled condition.
