Abstract

Carica papaya leaves are well known for their nutritional as well as medicinal value and have been used as therapeutic remedies for various diseases, such as dengue, malaria, and chikungunya. The project aimed to investigate the effects of different conventional plant extraction methods on the yield of bioactive compounds of Carica papaya leaves. Furthermore, the bioactivity of the leaves extracts also was investigated by using in-vitro free radical scavenging assay. Preliminary testing of the extract's toxicity was also conducted on HepG2 cells using the MTT assay. The presence of different types of bioactive phytochemical compounds within the leaves extracts was determined by several phytochemical screening. Phytochemicals such as saponins, alkaloids, fixed oil, phenolic compounds, flavonoids, terpenoids and cardiac glycosides which are potential antioxidants, are managed to be discovered in the methanolic extracts obtained through the reflux extraction method. Methanolic leaves extract derived from maceration showed the lowest cytotoxicity at the tested concentration range of 12.5 μg/mL to 200 μg/mL, as compared to the reflux and Soxhlet extraction methods, with the highest IC50 of 190 μg/mL. By conducting in vitro DPPH method, the free radical scavenging activity of the standard ascorbic acid was able to be determined. The percentage of radical scavenging activity of ascorbic acid is decreasing along with an increase of its concentration range from 4 μg/mL to 32 μg/mL, with EC50 of 17.50 μg/mL, which indicates its potency antioxidant properties
