Abstract
The aims of this project are to study the antioxidant and antimicrobial activities of the newly synthesized chalcone derived Schiff base and to determine whether the compound possesses synergistic effect as compared to its raw materials. In this project, a Schiff base with the IUPAC name of, (Z)-2-((E)-3-(4-hydroxyphenyl)-1- phenylallylidene)-N-methylhydrazine-1-carbothioamide, with the abbreviation of MTHC was synthesized via acid-catalysed condensation between 4-methyl-3-thiosemicarbazide (MT) and 4-hydroxychalcone (HC) in the equimolar ratio. The synthesized MTHC was characterized by melting point apparatus, Fourier transform infrared (FT-IR) spectroscopy, differential scanning colorimetric (DSC), thermogravimetric analysis (TGA), ultraviolet-visible (UV-Vis) spectrophotometer, mass spectroscopy (MS) and CHNS analyzer. Through the stated characterizations, it revealed that MTHC existed predominantly as the thione (C=S) tautomer in solid state. The antioxidant activity of MTHC was evaluated through DPPH and FRAP assays. Through these assays, it is found that MTHC exhibits the IC50 value of 8642 μM and FRAP value of 1.6683 mmol Fe2+/g. The antimicrobial activity of MTHC was also assessed qualitatively through disc diffusion assay. From the result, it showed that MTHC is inactive to inhibit the growth of the gram-positive bacteria, S. aureus and the gram-negative bacteria, E. coli.
