Abstract
The development of reliable, rapid, and cost-effective diagnostic test kits remains an important goal in biomedical research. Rapid diagnostic test kits such as Lateral flow assays (LFAs) are widely used due to their simplicity and flexibility. The sensitivity and stability of detection mechanisms, especially the antibody-conjugated nanoparticles are critical factors that influence performance of LFAs. The objective of this study is to explore quail egg-derived IgY as an alternative antibody source and highlights the possibility of AgNPs as a detection mechanism in LFAs. To achieve this, IgY was extracted from quail egg yolks using the water dialysis method, followed by purification via dialysis. AgNPs were synthesized through a chemical reduction process using sodium borohydride (NaBH4) and characterized for size and optical properties using UV-Vis spectroscopy and Fourier Transform Infrared (FTIR) Spectroscopy. Passive adsorption was carried out by mixing IgY with AgNPs optimized by ionic strength and incubation time to ensure stable IgY-AgNP conjugation. The conjugates were characterized using UV-Vis spectroscopy, FTIR and SDS-PAGE to confirm size, stability, and binding efficiency. Results showed successful conjugation of IgY to AgNPs, indicated by a red-shift in surface plasmon resonance (SPR) from 385 nm to 400 nm and increased the diameter in size. The conjugates remained stable under 4°C in the refrigerator. The results suggest that passive adsorption of quail-derived IgY to AgNPs has the potential to be used as a detection mechanism in the development of LFAs.
