Abstract
This research project focused on the extraction and characterization of bioactive compounds from S. crispa leaves, particularly highlighting the isolation and characterization of 2,5-di-tert-butylphenol (2,5-DTBP). The extraction process involved maceration with 80% ethanol and liquid-liquid partition, resulting in the successful isolation of the ethyl acetate (EA) extract with a yield of 0.91%. Phytochemical screening revealed the presence of alkaloids, phenols, and flavonoids, indicating the extract’s rich chemical composition. Further characterization using Fourier Transform Infrared (FT-IR) spectroscopy identified diverse functional groups within the EA extract. TLC profiling, FTIR, and Nuclear Magnetic Resonance (NMR) analyses were employed to characterize the isolated compound, 2,5-DTBP. Evaluation of antioxidant activity via the DPPH assay demonstrated significant potency for both EA extract (EC50 = 48.27 ± 3.33 μg/mL) and 2,5- DTBP (EC50 = 35.78 ± 0.78 μg/mL). However, the disc diffusion assay showed no inhibitory effects against Staphylococcus aureus and Escherichia coli at the concentration of 400 μg/mL to 50 μg/mL for EA extract and 200 μg/mL to 25 μg/mL for 2,5-DTBP. This research study facilitates future research on the isolation and identification of pharmacological agents. Understanding the therapeutic potential of these compounds, especially 2,5-DTBP with strong antioxidant properties, could pave the way for developing treatments to combat oxidative stress. Overall, this research highlights S. crispa leaves as a promising source of antioxidants for future investigation.
