Abstract

Cellulose biomass is the most abundant source to produce cellulosic ethanol as a renewable alternative energy source to petroleum. However, the production of cellulase has become the main limitation to cost-effective biofuel processing. Cellulolytic bacteria was isolated from cow dung and tested for clear zone formation around the colonies on the agar plates containing carboxymethylcellulose (CMC) as the sole carbon source. Out of 22 isolates, TJ02 showed the largest zone of hydrolysis. The crude enzyme was found to have a total enzyme activity of 0.049 mg glucose liberated / mg protein / 30 minutes. Isolate TJ02 was found to be Gram-positive with rod-shaped cells, positive in catalase, starch hydrolysis, triple sugar ion (TSI), and motility Tests. PH 7 and 72 hours incubation time were found to be optimized culture conditions for cellulase production. The isolate was assume as Bacillus spp. In the present study, cow dung was found to be a potential source to isolate cellulase- producing bacteria.
